Operative words
The Director shall impose such quarantine restrictions and regulations upon commerce and travel by railway, common carriers, or any other means, and upon all individuals as in his judgment may be necessary to prevent the introduction of communicable disease into the State, or from one place to another within the State.... C. No person shall interfere with any health authority having jurisdiction

What it required
Directs the Director (Secretary) of Health, on knowledge of a suspected case or outbreak, to 'take whatever steps necessary for the investigation and control of the disease' (sec. VII), to impose quarantine restrictions and regulations upon commerce, travel and all individuals (sec. X.A), bars local quarantine regulations of commerce or travel except by the Director's authority (sec. X.B), forbids any person to interfere with a health authority (sec. X.C), and imposes on attending physicians the duty to isolate (sec. IX); these are the rules whose violation 20-7-101 punishes and the source of the 'sole authority' recital in every 2020-2021 directive.
